Molecular Biology:

Biochemistry:

Expression of foreign proteins in E. coli and B. subtilis, protein extraction and purification, enzyme assay, zymogram analysis, agarose electrophoresis, SDS-PAGE, chromatography techniques.

Bacteriological Techniques:

Sterilization methods, isolation, screening and identification of bacteria; inoculation and culture of bacteria, yeast and fungi, competent cells preparation.

Bioinformation and Computer Skills:

Skillful at computer operating system and hardware, imaging compile (PowerPoint, Photoshop), webpage design and construction, web-based database searches and sequence analysis, and major software for molecular biology and data analysis such as BioEdit, BandScan, DNAstar and Primer Premier.

 

MAIN RESEARCH FIELDS INVOLVED

 

  • Bioenergy, cellulase characterization & engineering, cellulose hydrolysis mechanisms, high-level protein expression and extracellular secretion in E. coli and B. subtilis; metabolic engineering for producing secondary metabolites.
  • Investigation of the signal-less proteins secretion in bacteria. To elucidate the mechanisms of these proteins’ non-classical secretion and the relationship of their secretion with the pathogenesis. Development of the new target for the pathogentic bacteria’s control.
  • The establishment and application of the new molecular cloning and high-efficiency expression and secretion system in Bacillus subtilis strains; the recombinant expression and secretion, separation and purification of the proteins in Bacillus subtilis; development of new homologous recombinant vectors which can be generally used in gram positive bacteria easily.
  • Applied and environmental microbiology: the research of microbiological degradation of toxic organic pollutant. Isolation, screening and study of the characters of the bacteria strains from polluted soils and water for the biodegradation of organophosphorus pesticides; cloning and overexpression of the organophosphorus hydralase gene. The biodegradation and elimination of the pesticide residue from the contaminated environment and farm products.
  • The construction and application of the new promoter-trapping vectors with the mpd (methyl parathion degrading) gene or gfp (green fluorescence protein) gene as the reporter; cloning the strong promoter and signal peptide fragments for the high-level expression and secretion of the foreign proteins.
  • Plant pathogens biocontrol: Isolation, screening and identification of biocontrol bacteria strain from the collected soil samples for controlling the pathogenic fungi to the plant.
  • Characterization and performance comparison of several Saccharomyces cerevisiae.
